This MAb recognizes a protein of 36 kDa, identified as ICOS-L. It is a ligand for the ICOS receptor that initiates T and B cell proliferation and cytokine secretion. ICOS-L interactions play an essential role in T cell-dependent B cell activation in peripheral lymphoid organs such as spleen and lymph nodes. ICOS-L protein is present in myeloid leukocytes, and by Northern blot there are 2. 4, 3. 0, and 7. 0 kb transcripts in brain, heart, kidney, and liver, with lower expression in colon and thymus, and a 1. 1 kb transcript in leukocytes. Tumor necrosis factor alpha (TNF), granulocyte-macrophage colony-stimulating factor (GM-CSF) and interleukin-4 (IL-4) enhance its expression. The protein encoded by this gene is a member of the transmembrane 4 superfamily, also known as the tetraspanin family. Most of these members are cell-surface proteins that are characterized by the presence of four hydrophobic domains. The proteins mediate signal transduction events that play a role in the regulation of cell development, activation, growth and motility. This encoded protein is found in the asymmetrical unit membrane (AUM) where it can form a complex with other transmembrane 4 superfamily proteins. It may play a role in normal bladder epithelial physiology, possibly in regulating membrane permeability of superficial umbrella cells or in stabilizing the apical membrane through AUM/cytoskeletal interactions. Recognizes Nucleolin (NCL), the major nucleolar phosphoprotein of growing eukaryotic cells. NCL is located mainly in dense fibrillar regions of the nucleolus. It is found associated with intranucleolar chromatin and pre-ribosomal particles. Human NCL gene consists of 14 exons with 13 introns and spans approximately 11 kb. It has a predicted molecular weight of ~76 kDa with an apparent MW of 100-110 kDa, attributed to phosphorylation of its N-terminal domain. It induces chromatin decondensation by binding to histone H1. Recognizes a protein of 30 kDa which is identified as Prohibitin, an evolutionarily conserved protein with homologues found in yeast to man. It is located in the inner membrane of mitochondria. Although prohibitin mRNA and protein expression occurs throughout the cell cycle, maximum levels are detected during the G1/S phase transition and minimum levels are seen in S phase and the G2/mitosis boundary. Prohibitin is located exclusively in the mitochondria with the highest concentration on the inner membrane. Prohibitin is an ideal mitochondrial marker. Reacts with a protein of 57 kDa, identified as the L1 protein of human papilloma virus type 16 (HPV-16). It is the major capsid protein of HPV-16. Infection with specific types of HPV has been associated with an increased risk of developing cervical neoplasia. HPV types 6 and 11 have been associated with relatively benign diseases such as genital warts but types 16 and 18 are strongly associated with cervical, vaginal, and vulvar malignancies. The antibody reacts very strongly with formalin-fixed, paraffin-embedded tissues containing HPV-16 or -33; very weak reactions were occasionally observed with biopsy specimens or smears containing HPV-6 or HPV-11. CD45R, also designated CD45 and PTPRC, has been identified as a transmembrane glycoprotein, broadly expressed among hematopoietic cells. Multiple isoforms of CD45R are distributed throughout the immune system according to cell type. These isoforms arise because of alternative splicing of exons 4, 5, and 6. The corresponding protein domains are characterized by the binding of monoclonal antibodies specific for CD45RA (exon 4), CD45RB (exon 5), CD45RC (exon 6) and CD45RO (exons 4 to 6 spliced out). The variation in these isoforms is localized to the extracellular domain of CD45R, while the intracellular domain is conserved. Perilipin-2, also known as Adipophilin, belongs to the perilipin family, members of which coat intracellular lipid storage droplets. Recognizes a 33 kDa glycoprotein, identified as Nucleophosmin (NPM). It is predominantly localized in the nucleus of cells in most tissues. NPM is involved in ribosomal assembly and rRNA transport. It is an abundant protein that is highly phosphorylated by Cdc2 kinase during mitosis. This phosphoprotein moves between the nucleus and the cytoplasm. It is thought to be involved in several processes including regulation of the ARF/p53 pathway. A number of genes are fusion partners, in particular the anaplastic lymphoma kinase gene on chromosome 2. Mutations in exon 12 affecting the C-terminus of the protein are associated with an aberrant cytoplasmic location. Mutations in this gene are associated with acute myeloid leukemia. Recognizes a protein of 120-80 kDa, identified as E-cadherin. Cadherins comprise a family of Ca2 -dependent adhesion molecules that function to mediate cell-cell binding critical to the maintenance of tissue structure and morphogenesis. The classical cadherins, E-, N- and P-cadherin, consist of large extracellular domains characterized by a series of five homologous NH2 terminal repeats. Cytokeratins are a family of intermediate filament proteins that assemble into filaments through forming heterodimers of one type I Cytokeratins (Cytokeratins 9 to 23) and one type II Cytokeratins (keratins 1 to 8). CLP36, also known as PDLIM1, CLIM1 or Elfin, is a 329 amino acid cytoplasmic protein that associates with actin stress fibers at the cytoskeleton. Expressed at high levels in skeletal muscle and heart and at lower levels in colon, small intestine, spleen, lung, placenta, kidney, liver, thymus and pancreas, CLP36 functions as a cytoskeletal protein that is thought to act as an adaptor, bringing target proteins to the cytoskeleton. Specifically, CLP36 interacts with Clik1 (-actinin-1, thereby facilitating the association of Clik1 with actin stress fibers. CLP36 contains one PDZ domain and one LIM zinc-binding domain through which it conveys its protein-protein binding capabilities. This gene encodes the fatty acid binding protein found in epidermal cells, and was first identified as being upregulated in psoriasis tissue. Fatty acid binding proteins are a family of small, highly conserved, cytoplasmic proteins that bind long-chain fatty acids and other hydrophobic ligands. FABPs may play roles in fatty acid uptake, transport, and metabolism. Polymorphisms in this gene are associated with type 2 diabetes. The human genome contains many pseudogenes similar to this locus. FABP5 is also associated with poor survival in triple-negative breast cancer. This antibody recognizes a protein of 25-26 kDa, identified as Bcl-2. This antibody shows no cross-reaction with Bcl-x or Bax proteins. Bcl-2 suppresses apoptosis in a variety of cell systems including factor-dependent lymphohematopoietic and neural cells. It regulates cell death by controlling the mitochondrial membrane permeability. Bcl-2 appears to function in a feedback loop system with caspases. It inhibits caspase activity either by preventing the release of cytochrome c from the mitochondria and/or by binding to the apoptosis-activating factor (APAF-1). Expression of Bcl-2 inhibits programmed cell death (apoptosis). This MAb recognizes a protein of 56.5 kDa, identified as cytokeratin 10 (CK10). CK10 is expressed in all suprabasal layers of the epidermis. In the epidermis, expression of CK10 strictly parallels the extent of differentiation; it is absent in the basal layer, appears in the first suprabasal layers and increases in concentration towards the granular layer. However, CK10 is rarely detected in early stages of vulvar squamous carcinomas (tumors less than 2 cm, clinical stage I) regardless of the tumor grade. In larger and more advanced tumors (greater than 2 cm, clinical stages II and III), CK10 is detected very frequently.
